陕 西 中 医 学 院硕 士 学 位 论 文截 鼽 汤 对 过 敏 性 鼻 炎 大 鼠 骨 髓 中 EOS 及 血 中 IL-6 含 量 影 响 的 实 验研 究姓 名 ： 谭 艳申 请 学 位 级 别 ： 硕 士专 业 ： 中 医 五 官 科 学指 导 教 师 ： 张 雄20100401摘 要目 的 ： 本 实 验 借 助 现 代 医 学 科 学 的 研 究 手 段 和 方 法 ， 旨 在 观 察 截 鼽 汤 对 过 敏 性鼻 炎 的 疗 效 ， 并 从 其 对 过 敏 性 鼻 炎 大 鼠 骨 髓 中 嗜 酸 性 粒 细 胞 及 血 中 IL-6 含 量 的 影响 上 探 讨 其 作 用 机 理 ， 从 而 为 过 敏 性 鼻 炎 的 发 病 机 理 及 中 医 药 防 治 本 病 寻 求 有 力 的科 学 依 据 。方法：采用卵清蛋白为致敏原对 SD 大 鼠 进 行 过 敏 性 鼻 炎 的 模 型 复 制 ， 选 清 级健 康 SD 大 鼠 48 只 ， 随 机 分 为 空 白 对 照 组 、 模 型 组 、 西 药 治 疗 组 、 中 药 治 疗 组 四 组 ，每 组 12 只 。 观 察 并 记 录 SD 大 鼠 搔 鼻 、 打 喷 嚏 和 流 清 涕 等 鼻 部 症 状 ， 处 死 后 观 察 各组 SD 大 鼠 鼻 粘 膜 的 病 理 学 改 变 、 骨 髓 中 EOS 的 数 目 及 血 清 中 IL-6 含 量 的 变 化 。结 果 ： 1. 大 鼠 行 为 学 观 察 ： 治 疗 前 ， 模 型 组 、 西 药 治 疗 组 及 中 药 治 疗 组 鼻 部症 状 记 分 均 高 于 空 白 对 照 组 ， 差 异 具 有 显 著 性 意 义 （ P 0.01） ，说明 AR 大鼠模型复 制 成 功 。 治 疗 后 ， 西 药 治 疗 组 、 中 药 治 疗 组 鼻 部 症 状 记 分 均 低 于 模 型 组 （P 0.01）而 高 于 空 白 对 照 组 （ P 0.05） ， 说 明 中 药 、 西 药 治 疗 组 鼻 部 症 状 明 显 改 善 但 未 完 全恢 复 正 常 ； 西 药 治 疗 组 与 中 药 治 疗 组 间 差 异 不 明 显 （ P 0.05） ， 说 明 西 药 治 疗 组 与中 药 治 疗 组 在 改 善 AR 的 鼻 部 症 状 方 面 几 乎 等 同 。 2. 骨 髓 中 EOS 计 数 ： 模 型 组 大鼠 骨 髓 中 EOS 明 显 高 于 中 药 治 疗 组 （ P 0.01） ， 说 明 截 鼽 汤 能 够 有 效 的 降 低 AR 大鼠 骨 髓 中 EOS 的 作 用 。 中 药 治 疗 组 与 西 药 治 疗 组 比 较 ， 其 差 异 无 统 计 学 意 义 （ P0.05） 。 3. 血 清 IL-6 溶 度 的 变 化 ： 中 药 治 疗 组 大 鼠 血 清 中 IL-6 水 平 均 明 显 低 于 模 型组 （ P 0.01） ， 说 明 截 鼽 汤 能 有 效 的 降 低 AR 大 鼠 血 清 中 IL-6 水 平 。 中 药 治 疗 组 与西 药 治 疗 组 比 较 ， 其 差 异 无 统 计 学 意 义 （ P 0.05） 。结论：截鼽汤能够明显的降低 AR 大 鼠 骨 髓 中 EOS 及 血 清 中 IL-6 的 含 量 ， 有效 的 减 少 过 敏 性 鼻 炎 SD 大 鼠 搔 鼻 、 打 喷 嚏 的 次 数 ， 明 显 改 善 AR 典型的鼻部症状，从 而 达 到 治 疗 过 敏 性 鼻 炎 的 目 的 。关 键 词 ： 截 鼽 汤 ； 过 敏 性 鼻 炎 ； SD 大 鼠 ； EOS ； IL-6The Effect of Jie qiu tang on EOS in marrow and IL-6 in serumof Allergic rhinitis ratesAbstractObjective: The objective of the experiment is to observe curativeeffect of Jie qiu tang on rats with allergic rhinitis (AR) by modern medicineand scientific methods, and to discuss its treating mechanism from the effectof Jie qiu tang on EOS in marrow and IL-6 in serum of allergic rhinitis rates,to seek scientific evidence of treating and preventing the disease.Method: Establish allergic rhinitis models of SD rats by injectingvaccine (OVA Alumina polyvalent vaccine). Forty-eight healthy SD ratswere randomly divided into four groups: the normal group, the AR model group,the western medicine group, the traditional Chinese medicine (TCM) group.We will observing and recording the nasal symptoms such as scratching noses,sneezingand flowing mucus during the experiment. After killing the rats, observethepathological changes of the nasal mucosa of them in each group, and testthe expression of EOS in marrow and IL-6 in serum of rats with allergicrhinitis.Results: 1.Observation of nasal symptoms on the rats before and aftertreatment and marks show that: before treatment, the nasal symptoms marksof the AR model group, the western medicine group and the TCM group surpassto obviously the normal group and have significant differences（ P 0.01） .Itshow that allergic rhinitis models of SD rats are successful. after treatment,both the western medicine group and the TCM group were lower than the ARmodel group（ P 0.01） , but they surpass to the normal group（ P 0.05） .Itprove that both the western medicine group and the TCM group can improvethe nasal symptoms of the AR rats, but they can not cure it. they are almostequal between the western medicine group and the TCM group（ P 0.05） .2.The count of EOS in marrow: EOS in marrow of the AR model group surpassobviously to the TCM group （ P 0.01） .It prove that both Jie qiu tang canreduce effectively EOS in marrow of the AR rats. The western medicine grouphave no significant difference from the TCM group about reducing EOS（ P 0.05） .3.The level of IL-6 in serum: the TCM group are lower than theAR model group about The level of IL-6 in serum. It prove that Jie qiu tangcan reduce effectively IL-6 in serum. the western medicine group have nosignificant difference from the TCM group about reducing IL-6（ P 0.05） .Conclusion: Jie qiu tang can reduce effectively EOS in marrow and IL-6in serum of Allergic rhinitis rates. And it also can reduce times ofscratching noses and sneezing about the AR rats, obviously improving typicalnasal symptoms of the AR rats.
