荭草花水溶性化学成分的研究的论文李勇军,何迅,刘志宝,周雯,兰燕宇,王爱民,王永林【摘要】 目的研究荭草polygonum orientale花活性部位的化学成分。方法采用溶剂提取,应用色谱技术分离纯化,根据化合物理化性质和光谱数据鉴定其结构。结果分离并鉴定了6个化合物,分别为n-顺式-对羟基苯乙基阿魏酰胺(n-cis-feruloyltyramine, 1);n-反式-对羟基苯乙基阿魏酰胺(n-trans-feruloyltyramine, 2);n-p-香豆酰酪胺(paprazine, 3);5,7-二羟基色原酮(5,7-dihydroxychromone, 4);3,5,7-三羟基色原酮(3,5,7-trihydroxychromone, 5);5,7,4-三羟基二氢黄酮醇(5,7,4-trihydroxy-dihydroflavonol, 6)。结论化合物13为首次从该种植物中分离得到。【关键词】 荭草; 化学成分; 结构鉴定chemical constituents of flowers of polygonum orientaleli yongjun, he xun, liu zhibao, zhou wen, lan yanyu, wang aimin, wang yonglinschool of pharmacology, guiyang medical college,guiyang 550004, chinaabstract:objectiveto study the chemical constituents of the essential substance from the flowers of polygonum techniques were employed for isolation and purification of the constituents and their structures were determined by spectral analysis and chemical evidence. resultssix xxpounds were obtained and identified as n-cis-feruloyltyramine (1), n-trans-feruloyltyramin (2), paprazine (3), 5,7-dihydroxychromone (4), 3,5,7-trihydroxychromone (5), 5,7,4-trihydroxy-dihydroflavonol (6).conclusionxxpounds 1-3 were isolated from polygonum oriental for the first time.key words:polygonum oriental; chemical constituents; structure identification荭草花为蓼科植物荭蓼polygonum orientale l.的花序,又名水荭花,何草花,狗尾巴花;荭草花具有行气活血、消积、止痛的功效,用于治疗头痛、心胃气痛、腹中痞积等1。课题组在对荭草各部位药理筛选实验中发现荭草花水煮醇沉后的正丁醇萃取部位有较强的抗心肌缺血,减少心肌梗塞面积作用。为阐明其活性成分,作者对荭草花水煮醇沉后的正丁醇萃取部位进行了化学成分研究,在前期研究中已分离出9个化合物,在对其进一步的研究中作者又分离得到了6个化合物:n-顺式-对羟基苯乙基阿魏酰胺 (n-cis-feruloyltyramine, 1);n-反式-对羟基苯乙基阿魏酰胺 (n-trans-feruloyltyramine, 2);n-p-香豆酰酪胺 (paprazine, 3);5,7-二羟基色原酮 (5,7-dihydroxychromone, 4);3,5,7-三羟基色原酮(3,5,7-trihydroxychromone, 5);5,7,4-三羟基二氢黄酮醇(5,7,4-trihydroxy-dihydroflavonol, 6)。其中化合物13为首次从该种植物中分离得到,其结构式见图1。1 仪器与材料jeol ecx 500mhz核磁共振波谱仪,hp-lc-msd电喷雾质谱仪,vector-22红外分光光度计,agilent1100半制备高效液相色谱仪。色谱用硅胶(200300目)及硅胶g预制板为青岛海洋化工厂生产,聚酰胺(3060目)为浙江黄岩化工厂生产,c18反相硅胶为德国merck公司生产,sephadex lh-20为瑞士pharmacia biotech公司生产。实验样品采收自贵州省贵阳市鹿冲关,由贵阳医学院药学院生药学教研室龙庆德副教授鉴定为polygonum orientale l. 的干燥花序。2 方法与结果提取与分离取荭草干燥花序15kg,加水煎煮两次,滤液合并浓缩,加乙醇使含醇量达65%,静置过滤,回收乙醇,残留物用正丁醇萃取3次,回收正丁醇,残留物用乙醇溶解,上聚酰胺柱,用乙醇洗脱,收集乙醇洗脱液,回收乙醇得浸膏140 g。取浸膏用少量乙醇溶解,加150 g硅胶搅拌,晾干,研细,上样于1200 g硅胶(200300目)柱,用氯仿甲醇梯度洗脱,薄层检查,合并相同组分,再经反复硅胶柱色谱、sephadex lh-20柱色谱和半制备高效液相色谱纯化分离,得化合物 1 (680 mg),2 (1 g),3 (50 mg),4 (30 mg),5 (37 mg),6 (250 mg)。结构鉴定化合物1 白色粉末; esi-ms m/z: m+na+, mh; 1h-nmr ( dmso-d6, 500 mhz ) : (1h, br s, -oh), (1h, br s, -oh), (1h, t, j, =nh), (1h, s, h-5), (1h, d, j= hz, h-9), (2h, d, j= hz, h-4, 8), (1h, d, j= hz, h-8), (2h, d, j= hz, h-5, 7), (1h, d, j= hz, h-3), (1h, d, j= hz, h-2), (3h, s, 6-och3), (2h, m, h-1), (2h, t, j= hz, h-2); 13c-nmr ( dmso-d6, 125mhz ) : (c-1), (c-6), (c-7), (c-6), (c-3), (c-3, 4, 8), (c-4), (c-9), (c-2), (c-5, 7), (c-8), (c-5), (-och3), (c-1), (c-2)。经与文献2对照,确定化合物1为n-顺式-对羟基苯乙基阿魏酰胺 (n-cis-feruloyltyramine)。化合物2白色粉末; esi-ms m/z: m+na+, mh; 1h-nmr ( dmso-d6, 500 mhz ) : (1h, br s, -oh), (1h, br s, -oh), (1h, t, j, =nh), (1h, d, j = hz, h-3), (1h, d, j = hz, h-5), (2h, d, j = hz, h-4, 8), (1h, dd, j = , hz, h-9), (1h, d, j = hz, h-8), (2h, d, j = hz, h-5, 7), (1h, d, j = hz, h-2), (3h, s, 6-och3), (2h, m, h-1), (2h, t, j = hz, h-2); 13c-nmr ( dmso-d6, 125mhz ) : (c-1), (c-6), (c-7), (c-6), (c-3), (c-3, 4, 8), (c-4), (c-9), (c-2), (c-8), (c-5, 7), (c-5), (-och3), (c-1), (c-2)。经与文献2对照,确定化合物2为n-反式-对羟基苯乙基阿魏酰胺 (n-trans-feruloyltyramine)。化合物3白色粉末; esi-ms m/z: m+na+, mh; 1h-nmr ( dmso-d6, 500 mhz ) : (1h, s, -oh), (1h, s, -oh), (1h, t, j hz, =nh), (2h, d, j = hz, h-4, 8), (1h, d, j = hz, h-3), (2h, d, j = hz, h-5, 9), (2h, d, j = hz, h-5, 7), (2h, d, j = hz, h-6, 8), (1h, d, j = hz, h-2), (2h, m, h-1), (2h, t, j = hz, h-2);13c-nmr ( dmso-d6, 125mhz ) : (c-1), (c-7), (c-6), (c-3), (c-4, 5, 9), (c-4, 8), (c-3), (c-2), (c-6, 8), (c-5, 7), (c-1), (c-2)。经与文献3对照,确定化合物3为n-p-香豆酰酪胺 (paprazine)。化合物4淡黄色片晶; esi-ms m/z: m+h+, mh;1h-nmr ( dmso-d6, 500 mhz ) : (1h, s, 5-oh), (1h, s, 7-oh), (1h, d, j = hz, h-2), (1h, d, j = hz, h-8), (1h, d, j = hz, h-3), (1h, d, j = hz, h-6); 13c-nmr ( dmso-d6, 125mhz ) : (c-4), (c-7), (c-5), (c-9), (c-2), (c-3), (c-10), (c-6), (c-8)。经与文献4,5对照,确定化合物4为5,7-二羟基色原酮 (5,7-dihydroxychromone)。化合物5淡黄色针晶; ei-ms m/z: 194m+, 166, 152, 137, 124, 96; 1h-nmr ( dmso-d6, 500 mhz ) : (1h, s, 5-oh), (1h, s, 7-oh), (1h, s, 3-oh), (1h, s, h-2), (1h, d, j = hz, h-8), (1h, d, j = hz, h-6);13c-nmr ( dmso-d6, 125mhz ) : (c-4), (c-7), (c-5), (c-9), (c-2), (c-3), (c-10), (c-6), (c-8)。经与文献6对照,确定化合物5为3,5,7-三羟基色原酮(3,5,7-trihydroxychromone)。化合物6白色结晶; esi-ms m/z: m+na+, mh;1h-nmr ( dmso-d6, 500 mhz ) : (1h, s, 5-oh), (1h, s, 7-oh), (1h, s, 4-oh), (2h, d, j = hz, h-2, 6), (2h, d, j = hz, h-3, 5), (1h, d, j = hz, h-8), (1h, d, j = hz, h-6), (1h, d, j = hz, h-2), (1h, d, j = hz, h-3);13c-nmr ( dmso-d6, 125 mhz ) : (c-4), (c-7), (c-5), (c-9), (c-4),
